Fixed samples for fluorescence microscopy applications can take many shapes and forms, including cultured cells in microplates, multicellular spheroids, tissue on slides, and whole organisms. Depending on the type of sample and experimental question, different methods and processing steps are employed to optimize results. Identifying an appropriate fixation method is critical for preserving cellular and tissue structure. Effective staining methods require thoughtful consideration of permeabilization and blocking steps, antibody and counter stain selection, as well as incubation times and washing procedures.

Here we describe a number of general concepts that are essential for successfully conducting fixing and staining applications and provide specific tips and guidance for preparing a range of sample types.
