Monitoring Mitochondrial Status Using Confocal Imaging

Mitochondria are double membrane bound cellular organelles that generate most of the chemical energy necessary to power the cell’s biochemical reactions. These organelles use an electrical membrane potential across their membranes to drive oxidative phosphorylation and produce ATP.

Mitochondrial specific fluorescent dyes can be used to monitor membrane electrical potential, a key parameter in assessing mitochondrial status. Because their fluorescence brightness is proportional to membrane potential, the effect of agents that disrupt ion transport or increase proton permeability, such as the mitochondrial uncoupler FCCP, can be investigated using confocal imaging and analysis.
