Discovering New Ways to Advance Cell Biology Research with Lionheart FX

Creating an in vitro environment that is highly similar to that found in the body, including the presence of an extracellular matrix (ECM), is a critical aspect of reaping the most relevancy from cell-based research. Often, researchers find that optimizing culture environments is a time-consuming and costly process. The team at denovoMATRIX GmbH in Dresden, Germany, seeks to ease this burden by developing and manufacturing novel, pre-coated cultureware for specific cell types. Dr. Dejan Hušman,  CEO of denovoMATRIX,  explains that the specialized coatings used on the cultureware are composed of glycosaminoglycans and biomimetic peptides that can be combined to generate entire libraries of extracellular matrices for use in their user-customized myMATRIX and their flagship screenMATRIX products. Dr., Hušman  notes that development of the latter was dependent on their use of BioTek’s Lionheart™ FX Automated Microscope.

The versatile screenMATRIX is a 96-well culture plate containing a different biomimetic coating in each well. Using screenMATRIX, users can quickly identify an optimal microenvironment relative to their adherent cells, including stem cells, primary cells, cancer cells and immortalized cells, without the time and expense associated with testing separate ECM parameters. “When developing screenMATRIX, one challenge was to quickly and efficiently evaluate performance of the coatings in cell culture to eliminate the unsuitable coatings from those that were optimal,” Dr. Hušman recalls. Their existing manual fluorescence microscope could not keep pace with the sample throughput, and so they looked into a number of automated imaging solutions from various manufacturers. Lionheart FX was chosen as their preferred automated phase contrast and fluorescence imaging solution in part due to its comprehensive analysis software, small footprint, and responsive local technical support.

In addition to enabling development of the screenMATRIX, Lionheart FX reduces the amount of time and effort that denovoMATRIX cell biologists spend on imaging and analysis so that they can focus on high value activities to support the company’s growth. “Lionheart FX is incredible,” he says. “I am continually amazed that we can start with a screenMATRIX with fixed and stained cells, push a button, and return an hour later with reams of data, including beautiful stitched multichannel fluorescence images, heatmaps representing cell behaviors, and analyzed cell populations expressing a set of markers.”
